Leonard y Sheldon son dos físicos teóricos que comparten trabajo y apartamento. La serie comienza con la mudanza de Penny, su nueva y atractiva vecina, de la que Leonard se enamora desde el primer momento. La dificultad de los físicos para relacionarse con personas fuera de su entorno da lugar a situaciones cómicas.
Ambos son brillantes en su trabajo, pero tanto ellos como sus amigos Howard Wolowitz, un pseudo-galán sacado de una película psicodélica de los 60's, y Rajesh Koothrappali (de nacionalidad india) son representados como unos completos Geeks, muy alejados de las inquietudes de la gente corriente.
Rajesh tiene la incapacidad de poder hablar con mujeres, aunque esta discapacidad es inhibida por efectos externos como alcohol o estudios clínicos en un par de capítulos de la serie. Rajesh odia la comida India y se siente un poco discriminado por los demás.
Sheldon pasó del quinto año de primaria a la universidad. Ahora tiene dos PhD y un Máster. Sabe que su inteligencia es superior a la mayoría de la gente y lo saca a relucir en algunas situaciones. Aunque no lo necesita, como cuando se niega a ir a dar una conferencia el dice: "No necesito la aprobación de mentes inferiores". Sheldon tiene el Síndrome de Asperger, lo que influye en la mayoría de contestaciones, y de ahí que no sepa, como se ve en un episodio, lo que es el sarcasmo.

Leonard Leakey Hofstadter, (Johnny Galecki), Físico. Es un hombre de 22 años que no ha tenido mucho éxito con las mujeres, puesto que no sabe hablar con ellas. Sus amigos lo viven fastidiando de que morirá solo como ellos, pero él no se rinde y siempre trata de conquistar a su vecina Penny. Su mejor amigo es Sheldon, ya que aceptó vivir junto a él, le apasionan los cómics y los videojuegos, es un adolescente en el cuerpo de un hombre técnicamente. Ademas es intolerante a la lactosa.
Sheldon Cooper, (Jim Parsons), Físico. El mejor amigo de Leonard, del este de Texas. Tiene varios defectos de personalidad, como un pequeño caso de autismo, ineptitud social, tendencia a la vanidad, poca empatía, OCD y un severo caso de compulsión obsesiva. Siempre suele ir vestido con un suéter y encima una playera con logotipo de superheroe.
Penny (Kaley Cuoco), camarera. Es la única amiga del sexo opuesto del grupo. Es camarera en el famoso local de Cheescake Factory. Es el amor imposible de Leonard y salieron sólo por un episodio en la segunda temporada. Además está escribiendo una novela sobre su vida. No es muy inteligente pero sabe sacarse de apuros en contadas ocasiones. Sus relaciones siempre son malas, como ella las describe: "O sólo salgo con idiotas insensibles, o eran geniales y yo los convierto en idiotas insensibles".
Howard Wolowitz, (Simon Helberg), Ingeniero. Es una versión moderna de los Beatles. Siempre viste ropa ajustada y estrogodelica. Aún vive con su madre y su mejor amigo es Raj. Le gusta ir al apartamento de Leonard y Sheldon sólo por Leonard, ya que se demostró que no le tiene paciencia a Sheldon. Cree que todas las mujeres se sienten atraídas por el pero el caso es el contrario. En otras palabras, es un poco pervertido, pero de buenos sentimientos.
Rajesh Koothrappali, (Kunal Nayyar). Astrofísico. Es quien representa el típico estereotipo de un Indio en USA. Es quien menos problemas del grupo tiene salvo por uno, no puede hablar con las mujeres por una vergüenza patológica, ya que sólo les puede hablar si esta ebrio o si toma una droga especial.
Leslie Winkle, (Sara Gilbert), Física.Compañera de trabajo de Sheldon y Leonard. Es adicta al sexo, se siente atraída por Leonard y odia a Sheldon por su arrogancia.
